Homonuclear 1H-1H Gradient
Selective COSY Spectra
Homonuclear COSY experiment using gradient selection |
|
Details:
Pulse sequence: standard Bruker pulse sequence cosygs,
using pulsed field gradients
Pulsed Field Gradients: sine-shaped, 1 ms duration, maximum strength=20
G/cm;
gradients ratio= 1:1 for N-type coherence selection, spectra (a),
(c); 1:-1 for P-type coherence selection (b); no coherence selection in
(d); positions of the diagonal and antidiagonal peaks, coresponding to
P- and N-coherence selection, are indicated by green and blue diagonal
lines
PFG treatment methods: Coherence Transfer Pathways (a,b), Multilayer
('Salami') approach (c,d)
Spin system: three 1H nuclei at Ha=4.2
ppm, Hb=2.8 ppm, Hb'=3.4
ppm; scalar couplings: Jab=20 Hz,
Jab'=12 Hz, Jbb'=30
Hz
(this spin system corresponds to cysteine, the scalar couplings were increased
for illustration puproses)
Spectrometer frequency: 500 MHz (1H)
Acquisition: 256 t1-increments with 128 complex points (t2) each
Processing details: zero filling, apodization using 90o
shifted sinebell, magnitude mode processing
